An eligible Heroic Ula'tek kill contributes one qualifying boss toward the Raid section of the weekly Great Vault. Raid choices unlock after 2, 4 and 6 qualifying current-season boss kills.

This means an AotC-only run does not unlock the first Raid choice by itself when the character starts the week with zero qualifying kills. If you already have other eligible raid progress, the Ula'tek kill contributes to the next available threshold.
